报告摘要:
The development of drugs is highly regulated, since their quality, safety and efficacy must be demonstrated at every stage. Drug substances may contain impurities arising from synthesis, degradation products or, in the case of biologics, variants generated during production and storage. Their characterization and quantification are therefore essential to verify compliance with specifications, detect changes that could affect safety, and ensure batch-to-batch consistency. For decades, liquid chromatography has been widely used for pharmaceutical analysis and is considered a gold standard, thanks to its ability to separate the components of complex mixtures.
The pharmaceutical landscape is now changing rapidly. Alongside with small molecules, we have seen an increasing number of peptides and proteins, monoclonal antibodies, antibody-drug conjugates, fusion proteins, cell and gene therapies, and nucleic-acid-based medicines. These products are often much larger and structurally more heterogeneous, with closely related variants that can be difficult to distinguish. Chromatographic methods originally designed for small molecules therefore do not always meet current analytical needs. Chromatographic strategies must be reconsidered to improve speed and selectivity, facilitate mass spectrometry (MS) detection, and automate analysis.
This presentation will cover four complementary areas: accelerating the analysis of therapeutic proteins using very short columns; improving chromatographic selectivity to distinguish closely related molecular variants; Improving MS compatibility for native chromatographic modes; and developing online multidimensional methods coupled with MS for characterization of variants at different structural levels.
